SNATE-L

Artemether is used in combination with the Lumefantrine for effectively treats the resistant developed strains of falciparum malaria. Both the drugs are active against the uncomplicated species of malaria, Plasmodium falciparum and chloroquine-resistant strains. Artemether acts more rapidly i.e. it has rapid onset of action and clears rapidly out of the body thus it provides rapid relief from the malarial parasites by decreasing their number.
Combination of artemether and lumefantrine is used because it is better tolerated and have fewer adverse effects or produces less vomiting, no central toxicity.

Artemether belongs to the class of artemisinin derivative. It interferes with the red blood cells of the body.

Lumefantrine is the synthetic amino alcohol or derivative of fluorene and is belongs to schizonticide class. Lumefantrine is given only once a day.

  • In uncomplicated malaria
  • Chloroquine resistant malaria

Mechanism of Action-

Artemether is activated only after converting into its metabolite i.e. dihydroartemisinin which acts on the erythrocytic stages of plasmodium falciparum occurs in human beings by inhibiting the synthesis of nucleic acid and proteins.
Lumefantrine inhibits the protein synthesis by inhibiting the formation of Beta-hematin by preventing the complex formation of hemin.

Pharmacokinetics:

Artemether-
It is absorbed orally and its absorption increases with meals.
It shows 95% binding with serum proteins
It is rapidly metabolized in the liver
It is excreted via urine

Lumefantrine-
It is absorbed orally similarly its absorption increases with food
It is 97% bound to proteins
It metabolized in the liver
It is excreted via urine
