It is selective blocker of 5HT3 receptors that are located on the nerve terminals of vagus in the periphery and in the chemotrigger zone of the area postrema, Antineoplastic agents causes release of serotonin from the intestinal enterochromaffin cells which stimulates the vomiting center as well as receptors presents in area postrema.

It does not show activity against the dopamine receptors and other serotonin receptors.

  • Chemotherapy induced vomiting
  • Post-operative nausea and vomiting
  • Uremic pruritis
  • Radiation therapy induced nausea and vomiting
  • Cholestatic pruritis

Mechanism of action-

Ondansetron blocks the reflex produced by the enterochromaffin cells present in the small intestine thus prevents nausea and vomiting.

Pharmacokinetics-

Absorption- It is absorbed orally by GIT and undergoes first pass metabolism
Protein Binding- It is 73% bound to plasma proteins
Metabolism- It is metabolized in the liver
Excretion- It is excreted both via urine and feces
